چکیده

1. Highly purified arginineand lysine-rich histone was maleylated and then hydrolyzed with trypsin. A large amount of precipitate approximating 35% of the histone appeared during the demaleylation procedure. The precipitate was separated into four fractions on a Sephadex G-50 column and one fraction, tryptic peptide II from the maleylated histone, was found to be the COOH-terminal portion of the histone by analysis of residues released by carbox.ypeptidases A and B. 2. The sequence of the 48 residues in this fragment determined by sequence analysis and overlapping fragments of tryptic, peptic, and thermolysin digestion products is His-LeuGin-Leu-Ala-Ile-Arg-Asn-AspGlu Glu Leu Asn Lys Leu Leu-Gly-Lys-Val-Thr-Ile-AlaGln-Gly-Gly-Val-Leu-ProAsn-Ile-Gln-Ala-ValLeu Leu -Pro Lys Lys -Thr Glu Ser His-His-Lys-Ala-Lys-Gly-Lys.

Amino acid sequence and sequence variability of the amino-terminal regions of lysine-rich histones.

The amino acid sequence has been determined for the first 72 residues of a lysine-rich histone from rabbit thymus. These are the residues contained in a fragment released from the histone by treatment with N-bromosuccinimide. Peptides derived by tryptic, thermolysin, and chymotryptic digestion of this 72-residue fragment were used to reconstruct the total sequence. Analysis of the sequence reve...

Structural analysis of the glycine-rich, arginine-rich histone. II. Sequence of the half of the molecule containing the aromatic amino acids.

1. The glycine-rich, arginine-rich histone from calf thymus was hydrolyzed with chymotrypsin; the peptides produced were fractionated on Sephadex G-25. The peptides containing the aromatic amino acids were purified by rechromatography on Sephadex G-25 and Aminex A-4 columns and by peptide mapping. 2. The six aromatic amino acids of the glycine-rich, arginine-rich histone were accounted for in e...

Sequence homology and role of cysteine in plant and animal arginine-rich histones.

The two arginine-rich histones III and IV, isolated from pea buds and calf thymus glands, were separated by preparative disc electrophoresis and each was shown to be homogeneous by amino-terminal and carboxyl-terminal analysis. Homologous arginine-rich components from calf thymus and pea bud were found to be extremely similar or identical in their aminoand carboxyl-terminal amino acids, amino a...

Structural analysis of the glycine-rich, arginine-rich histone. 3. Sequence of the amino-terminal half of the molecule containing the modified lysine residues and the total sequence.

1. The amino acid sequence of the amino-terminal portion of the glycine-rich, arginine-rich histone (Residues 1 to 49) has been determined from studies on the tryptic, chymotryptic, and thermolysin peptides. This sequence overlaps with that of the carboxylterminal portion (Residues 50 to 102) defined previously to give the following sequence: acetylserGly-Arg-Gly-Lys-Gly-Gly-Lys-Gly-Leu-Gly-Lys...

Some Researches on Histones

The histone extracted from calf thymus glands is a complex system of proteins, which can be fractionated by chromatography on carboxymethyl cellulose columns into three principal fractions (1) very lysine-rich, (2) moderately lysine-rich, (3) arginine-rich. When examined by starch gel chromatography each of these gives more than one band. Methods have been devised for further separation of the ...
